چکیده

There is growing evidence that cell cleavage involves contraction in the region of the furrow and relaxation above the most distant parts of the spindle (see references 15 and 16) . The contractile mechanism has been likened to that of muscle, mainly as a result of studies of glycerinated models of dividing cells (7, 9) and extraction of an actomyosin-like material (8, 11) . Muscular contraction is activated by calcium ions, and various workers have implicated the Ca ion as a regulator of cell cleavage (15, 16) . We now report experiments designed to test this hypothesis. The results show that Ca ions are essential for cleavage ; but, at this stage, it is not possible to decide whether or not changes in the level of ionized Ca regulate cleavage as opposed to cleavage merely having a requirement for some ionized calcium .

Intracellular free calcium oscillations in normal and cleavage-blocked embryos and artificially activated eggs of Xenopus laevis.

We have measured levels of intracellular free calcium ([Ca2+]i) in albino Xenopus laevis embryos using recombinant aequorin and a photon-counting system. We observed sinusoidal oscillations in [Ca2+]i that had the same frequency as cleavage, with cleavage occurring when [Ca2+]i was lowest. An increase in calcium was seen to precede first cleavage. The cyclic changes in calcium were superimposed...

Integrin Linked Kinase (X-ILK) Function during Embryonic Development and within Adult Tissues of Xenopus laevis

Integrin linked kinase (ILK) is a serine/threonine protein kinase implicated in the phosphatidylinositol 3’kinase (PI3’K) pathway. Integrin linked kinase has been investigated in different organisms such as mammalian systems (human, mice, rat), insects (Drosophila) and nematodes (Cenorhabditis elegans), however to date little data regarding ILK research on amphibians has been reported. In...

Expression of hsp90 Alpha and hsp90 Beta during Xenopus laevis Embryonic Development

Background: Members of the eukaryotic Hsp90 family function as important molecular chaperones in the assembly, folding and activation of cellular signaling in development. Two hsp90 genes, hsp90 alpha and hsp90 beta, have been identified in fish and homeothermic vertebrates but not in poikilothermic vertebrates. In the present study, the expression of hsp90 alpha and hsp90 beta genes in Xenopus...

Calcium buffer injections delay cleavage in Xenopus laevis blastomeres

Microinjection of calcium buffers into the two-cell Xenopus laevis embryo delays cell division in a dose-dependent manner. Four calcium buffers in the BAPTA series with different affinities for calcium were used to distinguish between a localized calcium gradient regulating cleavage and the global calcium concentration regulating this event. DibromoBAPTA (Kd = 1.5 microM) was found to delay cle...

Morphological and morphometric study of early-cleavage mice embryos resulting from in vitro fertilization at different cleavage stages after vitrification

The aim of this study was to examine the possible morphological and morphometric changes resulting from vitrification of embryos at the cleavage stage. In this study, 30 mice early-cleavage embryos at different stages of cleavage, resulting from in vitro fertilization (IVF) techniques, were examined before and after vitrification. Digital images were taken from embryos before and after vitrific...
